Hepatoxicity mechanism of PFOS and PFOA using systems biology approach

Category: the CAS Knowledge Innovation Programs PI: Zhang Jie Execution time: 2011. 01- 2013. 12 This project intended to investigate hepatoxicity mechanism of PFOS and PFOA by using systems biology approach. The responses of L-02 cell lines to PFCs were investigated on the levels of mRNA, proteins and metabolites, and related sensitive and specific biomarkers were screened with high throughput. Mode of action of PFCs was established according to the identified biomarkers. Furthermore, the multi-dimensional data from transcriptomics, proteomics and metabolomics were integrated by using systems biology tools to unveil the complex interaction network between mRNA, proteins and metabolites, finally elucidated the hepatoxicity mechanism of PFCs on molecular level. This study can provide fundamental data and technical support to the risk assessment of PFCs.
