Study of microbial mechanisms of N2O emission in mangrove ecosystems

In this project, we select mangrove ecosystem as the research area of this study. The microbial mechanism of the N2O emission in the mangrove ecosystem will be studied by combining in situ field sampling with mesocosm experiments. The objects of this project are : 1) to measure the N2O emission of the samples ( both samples from the field sample and the mesocosm ) under different environmental conditions( salnity, substrate concentration and dry-wet cycles); 2) to quantify the functional gene copies by using the real-time PCR; 3)to reveal the key denitrifiers and to illustrate the microbial mechanism of the N2O emission, microbial communities of the total denitrifiers and the active denitrifiers will be compared by using denaturing gradient gel electrophoresis (DGGE). Research on denitrifying microbial community expression is gaining more and more attention now, and the study may reflect the denitrifying activity of the denitrifiers more directly and provide the evident and data support for the strategy for controlling the wetland N2O emission .
